Abstract: As faster alternatives to bioassay-guided fractionation, various affinity selection-mass spectrometry (AS-MS) methods have been developed and used for the isolation, characterization and identification of pharmacologically active compounds in complex natural product mixtures such as botanical and microbial extracts. The most successful of these approaches include size exclusion chromatography LCMS (SEC MS), pulsed ultrafiltration (PUF-MS), and magnetic microbead affinity selection screening (MagMASS). The throughput of these MS-based methods was enhanced over 100-fold by incorporating multititer well plate format, automated sample preparation, UHPLC, and automating data analysis using metabolomics software.
Our on-going collaboration with Douglas Feinstein at the University of Illinois at Chicago concerning rescue therapy for superwarfarin overdoses will be updated in view of the >200 poisonings and 7 deaths reported thus far in Illinois, Wisconsin and Maryland. In preclinical studies, UHPLC-MS/MS was used to investigate the metabolism and elimination of the superwarfarin rodenticide brodifacoum. In support of the 2018 poisonings caused by adulteration of illicit synthetic cannabinoids sold in multiple states as “K2” and “Spice”, we modified our UHPLC-MS/MS method to detect, identify and quantify multiple superwarfarins in serum from hospitalized victims.
